Scrubbing is basically an error-check on your pool and proactively looking for any data corruption. You definitely want these running routinely as well as SMART tests.

I think SMART is not running, does it has to do with the fact that I have 2 usb drives as boot?

"Normal" as in the defaults, but not "optimal."

Check your ESXi path selection policy, it's probably VMW_PSP_MRU for "Most Recently Used" - you want to switch to VMW_PSP_RR (Round Robin) which will cycle between all available paths:

And then change your RR IOPS limit from the default 1000 to a lower number (1 works, if you're wondering):

Could this config affect the performance of the "regular" NIC that users use to connect to the vm inside this esxi host? Should I do any special config? My server has 4 NICs, which I am using one for users and 2 for iSCSI, I still have one available.

No, this will only impact the storage traffic. Regular traffic should be set up with load-balancing or teaming depending on your switch capabilities.

Ok, I'm going to checkout the teaming for my my ESXi host
