I have dual Supermicro X10DRH-iLN4 motherboard with chassis SC826BE2C-R920LPB

Do I really need dual socket motherboard for 80TB data storage(Word,PDF,JPEG,MP4,video files) for 100 users ???

A 2nd CPU is unlikely needed, because FreeNAS performance is not CPU-driven but I/O-driven.
You can consider than even professional systems (TrueNAS X10 & X20) from iXSystems have only one socket .

Nevertheless a 2nd CPU may be interesting in the case you need to populate all the DIMM or PCIe slots of your motherboard.
