I have a Plex server on my TrueNAS.
After a few years, the HDD space is starting to run out.
Questions:
1. I have a second HDD where there is a lot of space, can I create a directory on the 2nd HDD e.g. "movie2" and add it to the plex server? How? Or
2. is it better to add a new HDD and add the whole drive to the Plex server? How?
Thanks.

You can add any number of library directories to Plex. But the better course of action, if possible, would be to expand your pool, either by replacing disks with larger ones, or by adding vdevs. It's hard to be specific since you haven't given any detail about how your storage is set up, but either is easy enough to do in TrueNAS.
