I'm working from home with a work mac given by my employer and I was wondering... If I login to the work vpn or have any hidden spy apps, can the hacker (in this case my boss?) access my local hard drives? Other devices? What can I do to prevent this from happening? Which setting should I check and change?

On the work PC, probably. On additional machines on your home network, that would require illegal activity by your boss and could be made much more difficult (can never say impossible) by using a different username/password (complex and long) from your work account and never accessing home resources from that work PC.

If you can, put the "work" Mac on it's own VLAN isolated from the rest of your network

And don't mix work and personal activity on same computer.
