So I am planning on building a (4) 2TB drive enclosure with Raid-Z. If I want to run time machine for a few macs, can I easily add this into a Raid-Z, and just give it like... 500gb? Or should it be in its own volume, separate from my (4) 2TB Raid-Z? Such as (1) 1TB for TM, and the (4) 2TB for everything else.

You should create a separate dataset within your volume for the AFP share. This is pretty easy to do.

I have the same disk setup - 4x2Tb, RAIDZ1.
All four disks are in one volume, Volume1.
I have one unlimited dataset "stuff", and one dataset "timemachine" that is set with a quota of 1Tb in the dataset options.
Each is shared by AFP, and the TM share is set to advertise itself as a TM destination in the AFP share options.

This works perfectly, although I have lately learnt that I'd be better off with 5 disks for performance reasons, see noobsauce80's presentation in the sticky at the top of the n00bs forum.
